Research Abstract
Over the last 5 years, the research conducted in my laboratory has centered on the physical mapping of human chromosome 5. Sequence Tagged Sites (STSs) were generated over the entire length of this 194 Mb chromosome. 303 new human STS markers were prepared and regionally ordered. These STSs were used to establish a Yeast artifical chromosome (YAC) framework map for the 50 Mb short arm(p) and other selected regions on the long arm(q) of chromosome 5. Special consideration was given to the region on the short arm where the critical regions (mental retardation and larynx) for the most common deletion syndrome, Cri-du-chat, have been localized (Goodart et al. 1994, Gersh et al. 1997). This integrated 5p map will serve as a resource for the the continuing localization of genes on the short arm of human chromosome 5 and will form the basis for construction of a high resolution E. coli based map for sequencing.
